I've noticed that it's hard to hear vocals on some songs. The quality is good, but the instrumentals overpower the vocals and it's difficult to hear everything else. Like I said, it's only with certain songs while other songs sound fine to me. I was wondering if it's better to adjust my amp settings or adjust the WinAMP equalizer (which is currently turned off).
What do you guys think?

You would probably get a better response in the car audio forum. But to answer your question... you need to adjust the eq. Your amp settings probably only allow for crossover and gain adjustments (maybe a bass boost too) to bring out the vocals you need to adjust the frequency bands.
Try to avoid relying on EQ with audio players because it tends to distort the overall sound. If I use EQ, it is to reduce the gain of certain frequencies - not increase. I'd rather leave it off.
If your sound card has a DSP of some sort, it will be more useful to use the EQ that comes with it to fine tune your system along with the built-in crossovers of the amp.
You want to adjust the amplifier gain so distortion is not apparent when maxing out the PC volume.

^ Just in case you don't have EQ adjustments on your sound card, or not see this under the volume control panel. I'll share this info anyhow:
1) Open up sndvol32.exe (that darn Windows volume control/mixer panel ahah).
2) Options => Advanced Controls (Enable)
3) Click on the Advanced button under the main volume slider. See if Bass/Treble is available. I'd use this as well for some final tweaks.
4) Press CTRL+S to get the compact version of the mixer window.
