Most likely the best way would be to install a metal panel that the board itself mounts to, then attach a grounding line from that panel (grounding plane) to your car's chassis. This will be the most effective way to ground your board and help eliminate buzz.

Your desktop doesn't buzz because the whole case is a grounding element, and the power noise actually finds its way back through the power supply and back to your house's grounding setup.

In a plexi case in a car with no grounding connections, there is no place for that excess power bleed to go, so you have to bleed it to the car's body/frame instead, which is how the manufacturer keeps your stock stereo from buzzing so badly.
