Hi, I think MacCar is the right place for this question.
Anyway, I can help a little, cause I had the same trouble on my macbook.
First of all, what GPS receiver do you use? most usb receivers use a USB-to-serial adapter in them, so you need the driver of that adapter.
My reciever is a BU-303 that requires the PL2303 drivers.
I had trouble installing the drivers I found on the net firs, then I found the latest version that works well in Mac OS X 10.4.10
Take a look at system profiler in the USB section to see if your receiver is detected (and write down manufacturer and product id if you don't know the model).
once you installed the drivers, you should have a new device in /dev/.
mine is /dev/tty.usbserial, so I have to start gpsdX with that device as parameter.
I don't know how route66 works, but probably you have to set the device path in the preferences.
